Early-lethal mutant identification through deep-sequencing of wild-type sibs

PI: Dr. Ian Moore (University of Oxford)

Description: This application proposes to explore the signature in wild-type sibs from the same F2 mapping cross. It is not currently possible to identify induced recessive mutations that result in embryo/seedlings lethal phenotypes, mainly due to the difficulty in extracting good quality or enough DNA. Early-lethal alleles will tend to appear in a lower frequency in the F2 wild-types sibs. The challenge is to establish the feasibility of this approach.
